When creating your UnitySync connection, you must specify that you want to create mail enabled contacts.
The default destination map template is ActiveDir-contact (mail enabled). This will create mail enabled contacts.
In an existing connection, you can view the templates in use by reviewing the General tab. If the destination template does ont include 'mail enabled', the connection was not created to create mail enabled contacts. You will need to create a new connection.
Use Connection>New to create a new connection. Be sure to select a Map Template of ActiveDir-Contact (Mail Enabled)
